The Trem2 R47H Alzheimer’s risk variant impairs splicing and reduces Trem2 mRNA and protein in mice but not in humans.

A recent publication from the lab of Christian Haass has shown altered splicing of mouse Trem2 that is dependent upon the presence of the R47H mutation and leads to a cryptic splice site in exon 2 and a deletion of 119 nucleotides.

The MODEL-AD project has similar data demonstrating the cryptic splice site.  We are currently using Western blots and whole-brain transcriptomic analysis to assay mutant protein levels.  For now, we argue that the conclusion that “functional data derived from Trem2*R47H knock-in mice cannot be translated to humans” put forward in this paper may be premature because full-length transcripts are generated in Trem2*R47H homozygotes, albeit at reduced levels.

In parallel, we are considering alternative strategies to create Trem2 alleles to model late-onset AD.

Workshop on the use of mouse models to study neurodegenerative disease

The Jackson Laboratory will present the 3rd annual Workshop on using mouse models to study human neurodegenerative disease in May.  This course will focus on the use of the innovative tools and methods, specifically in the laboratory mouse, for answering questions of neuron and circuit function and disease mechanisms during neurodegeneration.  The course will employ both didactic and hands-on instruction in both laboratory techniques and bioinformatics.

Center Manager position available

The Model Organism Development and Evaluation for Late-onset-Alzheimer’s Disease (MODEL-AD) project seeks a Center Manager for the Jackson Lab site in Bar Harbor, Maine.  MODEL-AD aims to create the next generation of mouse models for AD and develop new therapeutic targets. The Center Manager is an integral team member in this multi-project, multi-institutional grant.  The successful candidate will devote effort to managing the laboratory staff, overseeing mouse colony management, data production and analyses, and presenting research updates at internal and external meetings.  Administrative responsibilities include serving as liaison between investigators at JAX, Indiana University (IU), and University of California-Irvine (UCI), communications between JAX MODEL-AD project leaders, and managing documentation for data use and lab and animal protocols. The successful candidate will be able to apply for grants to develop their own research interests that leverage the resources being developed within MODEL-AD.
